Tangential flow filtration was tested for clarifying lemon juice for the food industry. Membranes with three different pore sizes were assayed: 100,000. Da (polysulfone), 0.20 mum (polypropylene), and 0.45 mum (polyvinylidene difluoride). Volatiles were extracted using a distillation technique and quantified by gas chromatography-mass spectrometry. The concentration of aroma compounds finally found in the clarified lemon juice (CLJ) increased with the membrane pore size and more apolar compounds were found in the CLJ obtained by using more hydrophobic membranes. The low concentration of terpene hydrocarbons of the lemon juices clarified using membranes of 100,000. Da and 0.45 mum made them ideal as acidulant agents in fruit preservation. On the other hand, the CLJ obtained using a membrane of 0.2 mum of pore size, with high recuperation percentages for terpene hydrocarbons (e.g. limonene) and aldehydes (neral and geranial), made this juice ideal for applications in which a lemon flavor is needed. One of the main results of this work lies in the fact that certain compounds reduce their concentration in a large percentage during the cross-flow filtration. This reduction is associated with its presence in the juice cloud and pulp whose are concentrated in the retentate. The elimination of these compounds is transcendental to use lemon juice as an acidifier because they are responsible for aromatic degradation in processed food, as, for example, canned peach halves in syrup. Therefore, the use of cross-flow filtration in the clarification allows the use of lemon juice as acidulant in food with very sensitive flavors to degradation during processing and storage.

Mutton is widely consumed in China, European Mediterranean countries, and other regions around the world and is expected to gain popularity as an alternative to pork in dry-cured hams. Changes in the physicochemical indices, volatile compounds, and endogenous enzyme activities during the processing of dry-cured mutton ham were investigated. Eighty-six volatile compounds were identified during the dry-curing process, with aldehydes being dominant. Oxidation values increased significantly during dry-curing and decreased during ripening (p <.05). The activities of lipolytic enzymes remained active throughout processing, although the activities of the enzymes decreased. Lipoxidase activity increased rapidly during the fermenting stage and then decreased gradually during ripening (p <.05). Superoxide dismutase activity increased throughout processing, whereas the catalase activity decreased. According to partial least square variable importance in projection analysis, 49 odor-related compounds were selected from dry-cured mutton ham as specific compounds which are related to lipid oxidation and endogenous enzyme activities. Heatmap shows that the volatile compounds were correlated with enzyme activity and the oxidation value of dry-cured mutton ham during processing. Practical applications: Xinjiang dry-cured mutton ham has an intense and a long-lasting aroma. Understanding the mechanism of aroma formation related to lipid oxidation could help us to improve the product quality and optimize the duration of processing. Therefore, the present work identified the characteristic flavor compounds and provided the changing regularity of these compounds at various stages to facilitate the generation of them. The relationship between susceptibility of different peach cultivars (cvs) to the Mediterranean fruit fly (medfly), Ceratitis capitata, and the volatile composition of ripe fruit of each cv has been investigated, since understanding the fruit-insect interaction mechanism is crucial for developing control strategies for such a pest. Among the volatile compounds detected, 88 could be identified. The main differences found in the volatile composition of the cvs, concerned the relative abundance of esters. The least susceptible cvs, above all Percoca Romagnola 7, contained the higher amounts of hexenyl, hexyl, 3-methylbutyl, butyl and 2-methylpropyl esters; among these, some C6 derivatives detected, such as (Z)-3-hexenyl acetate, are known to act as priming agents, enhancing plant defence response to insects. Instead, a lower relative content of methyl esters, such as methyl hexanoate and methyl octanoate, known to act as medfly pheromone and attractant respectively, was found in the least susceptible cvs.

Purpose: Sclerotiorin, an azaphilone produced by some filamentous fungi including Penicillium sclerotiorum, is a pigment with variety of biological activities including lipoxygenase inhibition, reduction of cholesterol levels, and anti-cancer properties. Sclerotiorin has potential use in pharmaceutical as well as food industries. In this context, the purpose of this study was to provide a simple and robust procedure for optimised production of sclerotiorin by P. sclerotiorum using a central composite design developed through response surface methodology (RSM) and to identify the molecule(s) involved in the signalling mechanism in P. sclerotiorum. Methods: The optimisation of sclerotiorin production was carried out using RSM in shaken flasks and the obtained results were then replicated using a 2-L stirred tank bioreactor. Penicillium sclerotiorum ethyl acetate culture extract was analysed using thin layer chromatography (TLC) and potential signalling molecules were identified using Gas chromatography-mass spectrometry (GC-MS). Results: The experimental studies suggested an increase in the sclerotiorin production by 2.1-fold and 2.2-fold in shaken flasks and stirred tank bioreactors respectively. Further analysis of P. sclerotiorum ethyl acetate culture extract reported the presence of ricinoleic acid, an oxylipin, belonging to a family of signalling molecules tentatively involved in the enhancement of sclerotiorin production. Conclusion: This paper has highlighted the positive effect of the optimal supplementation of P. sclerotiorum culture extracts for enhanced production of sclerotiorin. It has also examined potential molecules involved in the signalling mechanism in P. sclerotiorum culture extract for the overproduction of sclerotiorin.

A dynamic headspace sorptive extraction (DHS) combined with thermal desorption (TD) and coupled with gas chromatography-mass spectrometry (GC/MS) was developed for the determination of 11 esters which contribute to the fruity aroma in sweet wines. A full factorial (4 factors, 2 level) experiment design was used to optimize the extraction conditions and the results were evaluated by multiple linear regression (MLR) and principal component analysis (PCA). The esters showed optimal extraction using an extraction temperature of 30 C during 20 min, and a subsequent purge volume of 300 mL and dry volume of 50 mL. Afterwards, quantification was achieved using calibration curves constructed for each ester with linear regression equations having correlation coefficients (R2) ranging from 0.9894 to 0.9981. The proposed method was successfully validated and showed good intermediate precision, repeatability and accuracy values for all the monitored compounds. Finally, the method was applied to quantify esters, with fruity aromatic notes, of sweet white and red wines, elaborated with different winemaking processes.

Through extensive solvent screening, acetonitrile was identified as the best solvent for solution-based crystallisation, due to its capability to produce compact parallelepipedic crystals and its comparably wide cocrystal phase region. Crystallisation conducted at 60 C was found to increase the conversion rate and the material efficiency compared to room temperature. By application of an advanced seeding procedure, high-quality cocrystals in the size region of 180?250 mum were produced in laboratory-scale antisolvent and cooling crystallisations. By pilot-plant-scale batch reaction cocrystallization, cocrystals with a volume-weighted mean diameter of 33 mum were produced in quantities of 250 g per day.

Among the 28 compounds studied, only 2-benzoxazolinone, 2-indanone and gamma-valerolactone showed similar inhibitory activity in both species. Indan had a lower IC50 for human than for mouse coumarin 7-hydroxylation, whereas the IC50 of 24 other compounds was higher for human than for mouse coumarin 7-hydroxylation. 5. The largest difference in IC50 between mouse and human activity was observed with 5-substituted phenyl, pentyl, hexyl, heptyl or octyl gamma-lactones or 6-substituted delta-lactones. IC50 of gamma-undecanolactone and gamma-decanolactone was 500 times lower for mouse than human coumarin 7-hydroxylation. 6. The difference in the IC50 between human and mouse coumarin 7-hydroxylation decreased substantially with the corresponding compounds without the lactone ring. 7. It is concluded that certain 5- or 6-position substituted gamma- and delta-lactones are potent inhibitors for mouse CYP2A5 but not for the orthologous human CYP2A6 and that the active site of CYP2A6 could be smaller than the active site of CYP2A5.

The possible impact of genetically engineered plants that degrade the quorum sensing (QS) signal of the plant pathogen Pectobacterium carotovorum was evaluated on non-target plant-associated bacterial populations and communities using Nicotiana tabacum lines expressing the lactonase AttM that degrades QS signals (AttM), and the wild type (WT) parent line. Cell densities of total culturable bacteria and those of selected populations (pseudomonads, agrobacteria) isolated from plant rhizospheres and rhizoplanes were comparable whatever the genotype of the plants (AttM or WT). Similarly, cell densities of members of the bacterial communities relying upon acyl-homoserine-lactones (AHLs) to communicate, or naturally degrading AHL signals, were identical and independent of plant genotype. Bacterial populations isolated from the two plant genotypes were also analyzed irrespective of their culturability status. DGGE analyses targeting the rrs gene (16S rRNA gene) did not reveal any significant differences within these populations.
