The TOPol. Sub-Structural Mol. Design (TOPS-MODE) approach has been used to address the adenosine kinase inhibitory activity of pyrrolo[2,3-d]pyrimidine nucleoside analogs. A model capable of describing around 77% of the variance in exptl. activity of 32 analogs was developed on the basis of this approach. In contrast, out of nine different approaches, including the use of Constitutional, Topol., BCUT, 2D autocorrelations, geometrical, RDF, 3D Morse, WHIM, and GETAWAY descriptors none were able to explain more than 70% of the variance in the above-mentioned property with the same number of descriptors. Although statistically significant models were derived containing descriptors other than spectral moments, the model that exhibited the best fit was based on these variables.
