Synthesis and biological activity of alpha-methylene-gamma-lactones as new aroma chemicals

Seven kinds of alpha-methylene-gamma-lactones with an alkyl group at the C-4 position were synthesized according to a previously described method, with yields of 28-34%. These alpha-methylene-gamma-lactones had characteristic and unique odors. All alpha-methylene-gamma-lactones added a roast-like odor to materials. The antimicrobial effects of alpha-methylene-gamma-lactones were investigated by using a paper disk diffusion method. The results showed the alpha-methylene-gamma-lactones inhibited the growth of three bacteria (Staphylococcus aureus, Escherichia coli, and Pseudomonas fluorescens) and two fungi (Saccharomyces cerevisiae and Aspergillus niger). In particular, alpha-methylene-gamma-undecalactone and alpha-methylenegamma-dodecalactone exhibited potent inhibition of the growth of these microorganisms compared to butyl p-hydroxybenzoate as standard antibiotic. The umu test revealed that the alpha-methylene-gamma-lactones suppressed the SOS-inducing activity of three mutagens, furylfuramide, UV irradiation, and TrpP-1, respectively. The antimicrobial effects and the suppressive effects of SOS induction by alpha-methylene-gamma-lactones had a tendency to intensify as the number of carbons in the side chain increased.

A convenient and high yielding method for the cleavage and scavenging of p-methoxybenzyl protecting group of several alcohols using tert-butyl bromide in refluxing acetonitrile is described. Application of off-line pyrolysis with dynamic solid-phase microextraction to the GC-MS analysis of biomass pyrolysis products

Pyrolysis coupled with dynamic solid-phase micro extraction (Py-SPME) followed by GC-MS analysis was applied to the determination of volatile compounds evolved by a micro-scale off-line pyrolysis apparatus, in order to extend the information affordable with this type of analytical equipment. The Py-SPME method with a carboxen/PDMS fiber working in the retracted mode was tested on four biomass samples (switchgrass, sweet sorghum, corn stalk and poplar) for qualitative analysis of semi-volatile pyrolysis products and quantitative determination of main volatiles (C1-C4) pyrolysis products. The developed procedure allowed capturing and analysis of all GC analyzable compounds, without memory effects and with good peak resolution also for early GC-eluting compounds. Twelve main volatile pyrolysis products, including hydroxyacetaldehyde and acetic acid, were successfully quantified; in spite of the intrinsic variability introduced by dynamic SPME sampling, results were relatively accurate and consistent with literature data on bench pyrolysis reactors.

Dehydrogenative alpha-oxygenation of ethers with an iron catalyst

Selective alpha-oxidation of ethers under aerobic conditions is a long-pursued transformation; however, a green and efficient catalytic version of this reaction remains challenging. Herein, we report a new family of iron catalysts capable of promoting chemoselective alpha-oxidation of a range of ethers with excellent mass balance and high turnover numbers under 1 atm of O2 with no need for any additives. Unlike metalloenzymes and related biomimetics, the catalyst produces H2 as the only byproduct. Mechanistic investigations provide evidence for an unexpected two-step reaction pathway, which involves dehydrogenative incorporation of O2 into the ether to give a peroxobisether intermediate followed by cleavage of the peroxy bond to form two ester molecules, releasing stoichiometric H2 gas in each step. The operational simplicity and environmental friendliness of this methodology affords a useful alternative for performing oxidation, while the unique ability of the catalyst in oxygenating a substrate via dehydrogenation points to a new direction for understanding metalloenzymes and designing new biomimetic catalysts.
